چکیده
Structured illumination microscopy (SIM) is widely applied due to its high temporal and spatial resolution imaging ability. sCMOS cameras are often used in SIM their superior sensitivity, resolution, field of view, frame rates. However, the unique single-pixel-dependent readout noise may lead reconstruction artefacts affect accuracy subsequent statistical analysis. We first established a nonuniform model address this issue, which incorporates offset, gain, variance based on process. The simulation indicates that pixel-dependent causes reconstructed superresolution (SR) image. Thus, we propose novel noise-corrected algorithm derived from model, can effectively suppress noise-related improve signal-to-noise ratio (SNR).

Superresolution Structured Illumination Microscopy (SR-SIM)
How Superresolution with ELYRA S.1 works The resolving power of a light microscope is limited to approximately 200nm in the lateral (XY) and 500nm in the axial (Z) direction. This so-called diffraction limit has been fi rst described by Ernst Abbe in 1873 (reference 1) and still holds true today. In simple terms, it posits that objects cannot be resolved with a conventional light microscope if ...
متن کاملQuantitative sectioning and noise analysis for structured illumination microscopy
Structured illumination (SI) has long been regarded as a nonquantitative technique for obtaining sectioned microscopic images. Its lack of quantitative results has restricted the use of SI sectioning to qualitative imaging experiments, and has also limited researchers' ability to compare SI against competing sectioning methods such as confocal microscopy. We show how to modify the standard SI s...
متن کاملOTF compensation in structured illumination superresolution images
The lateral resolution of an imaging system is limited by its numerical aperture and the wavelength. Structured illumination incident on the object heterodynes the higher spatial frequencies of the object with the spatial frequency of the sinusoidal illumination into the passband of the imaging system providing lateral superresolution. This idea has been implemented in microscopy. Multiple imag...
متن کاملA joint Richardson—Lucy deconvolution algorithm for the reconstruction of multifocal structured illumination microscopy data
We demonstrate the reconstruction of images obtained by multifocal structured illumination microscopy, MSIM, using a joint Richardson–Lucy, jRL-MSIM, deconvolution algorithm, which is based on an underlying widefield image-formation model. The method is efficient in the suppression of out-of-focus light and greatly improves image contrast and resolution. Furthermore, it is particularly well sui...
متن کاملPlasmonic structured illumination microscopy.
We propose a super resolution imaging technique called plasmonic structured illumination microscopy (PSIM), which combines the structured illumination microscopy technique with the tunable surface plasmon interference. Because of the high-resolution enabled by using surface plasmon interference as an illumination source, PSIM possesses higher image resolving power compared with conventional str...
